Unstructured mesh to structured mesh

I am now working on Ansys 14.5 with Fluent. I don't know how to transform unstructured grid/mesh to structured grid/mesh . if anyone can suggest me about that , it will be a great help for me

you are talking about hexa mesh?

For a single block structured grid, Gambit naturally produces a cell numbering which is structured. You just need to figure out, when cycling over the cells, which of the i,j,k indices (respectively along x, y, and z direction) varies faster, then the second one, etc.
